The 652-square-metre block on Anna Catherine Drive is listed for $630,000.
Stockdale and Leggo Port Fairy agent Sarah McCorkell said there were very few blocks with sea views available.
"It is becoming the most expensive block over on South Beach without absolute ocean frontage for sale due to limited land available in Port Fairy as a whole," Mrs McCorkell said.
"There are still a few blocks scattered along Ocean Drive with ocean views.
"However, they have been long-held blocks and may not come up for sale if the current owners have plans to develop them for themselves. Opportunities to build with ocean views is starting to dwindle as each block becomes developed."
Mrs McCorkell said the block had only been listed on Tuesday night and there was high demand for Port Fairy land.
"I think it is commonly known that all along the coast there has been a huge rise in prices and demand for properties in coastal towns," Mrs McCorkell said.
She said there had been a lot of speculation about Melbourne buyers wanting to invest in the town.
However, she believes the majority of buyers are from the district.
"Port Fairy has always been a choice destination for farmers," Mrs McCorkell said.
"They can get away and have a break without having to travel too far should they need to get back in a hurry to the farm. And long-term it's been a popular choice for retirement. So when farmers are doing well - which they have been - it tends to influence prices in town.
"On the flip side we have also seen a lot of young families relocating to Port Fairy and it's been lovely to see plenty of new faces and families joining schools and daycare."
Mrs McCorkell said central house prices were also attracting record prices.
"Again it comes back to the lack of supply and high demand," she said.
