Brown is the only Australian woman in the time trial.
In the road race she will be joined by Mitchelton-Scott teammates in Spratt, Sarah Roy and Lucy Kennedy.
Brodie Chapman, Shara Marche (FDJ Nouvelle Aquitaine Futuroscope) and Rachel Neylan (Cronos Casa Dorada Women Cycling) are also in the team.
This year's championship offers challenging road race routes which will see the women will tackle 2750 metres of climbing over their 144km course.
The time trial course will feature a flat 32km circuit set to favour specialists of the race against the clock.
The reliable Brown, who is making her second appearance in the grand tour, is set to play a vital role as well as giving the team options across the nine days of racing.
The former Australian time trial champion will share domestique duties with Kennedy, Roy and Dutch rider Moniek Tenniglo.
Brown enters the tour in promising form after finishing as the top Australian rider in La Course by Le Tour.
The Giro Rosa, which is the only women's grand tour on the calendar due to the coronavirus pandemic, begins on Friday, September 11 with a team time trial.
Mitchelton-SCOTT Sports Director Alejandro Gonzalez-Tablas said versatility would help the team achieve its goals.
"We bring a good mix of riders to work either on the flat or on the mountains," he said.
"In a race like this, the team is what makes the difference and I believe we line up with a strong team to support our leaders."
