Gak Thuk, 15, and his 11-year-old brother Magok Thuc were last seen by their family leaving for school at about 9am on Friday.
Magok is believed to have attended a school sports day on Friday but his brother did not turn up.
Friends reported seeing Gak at a shopping centre in Melton on Saturday, a police spokesman said.
Both boys were last seen wearing white t-shirts and black jackets. They are known to frequent the Melbourne CBD.
"Police believe that the boys are together but have concerns for their welfare due to their age and the length of time that they have been missing," the spokesman said.
They have released pictures of Gak and Magok in the hope it will assist in locating them.
Anyone who sees either boy is urged to contact Melton Police on 9747 7999.
