Threlfall has coached the Seagulls for the past two seasons having previously served as an assistant coach at Koroit.
Port Fairy Vice-President Shaun Murrihy said the club was thrilled to have Threlfall again at the helm.
"Appointing Shane for a third year gives the club great stability and a great platform to move forward," Murrihy said.
"I know Shane was disappointed with the result this year but if you look at the big picture it was a tough year.
"We had some injuries to key players early on and I think we also may have underestimated some of the other teams in the competition.
"Shane has got the respect of everyone at the club and we are all very happy going forward with him as the club coach."
The Seagulls have also re-appointed Tom Taylor as reserves coach and Paul Butters as coach of the Under 18's.
Threfall said the club had already started recruiting players for next season with a long wish list of players he was keen to see in the Seagulls colours next season.
"We will start trying late October of early November and we will be trying to get a competitive team together for our supporters" Threfall said.
Ruckman and 2010 bets and fairest winner Tim Robertson will not be with the Seagulls next season as he will return to his home club of Finley.
Centre half-forward Rory Mansell has returned to Tasmania and also looks unlikely to make it back to Port fairy next season.
The rumour mill has been busy on player movement from Port Fairy with Threlfall questioned on a number of these.
Threlfall said key onballer Brad Thomas would be with the Seagulls in 2011 despite speculation he was considering a return to Hamilton.
He said the club was talking with Jason McIntosh about his plans for next season while rumours Trinity Handley may be on the move was something Threlfall said he was unaware of and that he believed Handley would be at the club next season.
Forward Jet Dowie has been linked to a Warrnambool and District league cub but uncertainty remains about where he will play in 2011 with Port Fairy believed to still be an option.
