Tralee Rose, with top jockey Craig Williams aboard, finished less than 1.5 lengths from the winner No Effort in the 1700-metre race.
Trainer Symon Wilde said Tralee Rose, who is on a Caulfield and Melbourne cup, path ticked a lot of boxes with her first-up performance.
"We've come away very happy," Wilde told The Standard.
"Tralee Rose did exactly what I wanted her to do. She finished the race off strongly.
"She's going to be a lot better over more ground. I think she'll really appreciate running in the Naturalism Stakes which is over 2000 metres in three weeks.
"We'll give her a freshen up to have her ready for the Naturalism - if you win the Naturalism you get a golden ticket into the Caulfield Cup.
"The Bart Cummings is also on the agenda, a win in that race would get her a run in the Melbourne Cup.
"Willo gave us a glowing report after his ride on Tralee Rose."
Tralee Rose won the Bagot Handicap and the Lord Reims Stakes before being unplaced in the Adelaide Cup in her last campaign.
Meanwhile, imported mare Turaath scored her third consecutive metropolitan win for Warrnambool trainer Maddi Raymond in a $130,000 restricted mares race.
Turaath hung on to defeat Annavisto in the 1400-metre race.
Raymond said after the win Turaath was a tough horse.
"Turaath fought it out to the line strongly," she said.
"She's had a big preparation, we'll just take her home and see how she recovers from the run before working out what we do with her next."
Turaath has won four races from her 12 starts.
