Detective Senior Constable Richard Hughes, of the Warrnambool police crime investigation unit, said the incident happened between 1.30am and 8am.
He said a semi-automatic 22-carbine Sturm Ruger rifle was stolen from the front passenger seat of a vehicle parked in the driveway of a private residence in north Warrnambool.
The firearm was locked in a case which was chained to the seat and concealed.
Detective Senior Constable Hughes said there was no forced entry to the vehicle but the case was prised open.
"To have illegally obtained firearms circulating in the community is deeply concerning," he said.
"They can only be used for nefarious purposes.
"Anyone with any information is strongly urged to contact the Warrnambool police CIU on 5560 1153."
