The race was set to go ahead but the coronavirus pandemic forced the organisers to cancel the cycle team relay at the last minute.
President Peter Bollard said last year's cancellation weighed heavily on the committee's decision to run it on March 27-28 this year.
"Last year at the last minute we had to cancel and refund all the fees and we had more than 800 riders so it was an effort to refund," he said.
"We were weighing up whether to run it or not with COVID but (virus) numbers have been slow or non-existent and if it was a nice, fine, sunny day with north winds and we had already cancelled we would be kicking ourselves.
"So we thought if we go as if the event is going ahead and if something happens we can always cancel it if we have to like last year."
Mr Bollard said the Murray to Moyne was a tier three event and organisers had been working hard to make it a COVID-safe event.
The Murray to Moyne helps raise funds for hospitals and heath services across the country and also continue the dream of Graham "Woody" Woodrup's push of getting people to realise the benefits of riding a bike.
Entries are open until Wednesday. Bollard said team entries were significantly down on previous editions due to the impacts of the pandemic on fundraising events.
The event has four different starting points at Mildura, Swan Hill, Echuca and Hamilton.
The popular night riders entry travels from Mildura, Swan Hill or Echuca into Hamilton on the Saturday night and then into Port Fairy on Sunday morning.
The day timers ride 250-300km on Saturday until dusk, skip the night ride to Hamilton and then ride 90km on Sunday morning to Port Fairy.
Bollard said the Sunday 90-kilometre ride from Hamilton to Port Fairy had been a popular ride this year.
