Health Minister Martin Foley made the announcement on Saturday morning at the Royal Melbourne Hospital and said the blitz by the Department of Justice and Community Safety would focus on people maintaining social distancing and wearing masks.
"We will be rolling out in many different settings over the course of the weekend a blitz of compliance with businesses, be they hospitality, be they venues where people gather," he said.
Mr Foley said police, authorised officers from the Department of Health and Human Services and WorkSafe inspectors would target workplaces considered to be high risk.
"We need to make sure that the enormous sacrifices and the enormous progress that Victorians have made over the course of the pandemic are sustained," he said.
"And that means everyone doing the right thing.
"And that's why every business, every workplace, everyone who is out there in the community has a role to play to make sure that we stay open and that we never see the situation where we've seen here at the Royal Melbourne and other places where we see those infection levels rise again."
Mr Foley also announced $9.8 million funding to ensure wellbeing support for the healthcare workforce.
"During the course of the pandemic health care workers have stood up like never before," he said.
"Whether they be... the orderlies, the security guards, the cooks, the cleaners as well as the professional staff... the nurses, the pathologists, the doctors, the ambos."
