It comes after COVID-19 positive tests in Port Fairy two weeks ago put training on the back-burner.
Club presidents Steve Hoy (Koroit) and Noel Black (Port Fairy) said they had gathered information and were comfortable with the decision.
But both said plans could be subject to change.
"I suppose last week we had some initial concerns," Hoy said.
"But Port Fairy have been great throughout with their communication and they've had their finger on the pulse with what's going on in the town and getting great information.
"We surveyed our (players') parents, we met as a committee and we're prepared to go.
"If something happens as late as Friday or Saturday, that's the world we live in."
Black said the Seagulls had kept abreast of the COVID-19 situation in Port Fairy and based their decision on information available.
"With the four people who were a-symptomatic positive, there doesn't seem to be any follow-up transmission around the area so we believe it's fine to play," he said.
"After difficult times due to the COVID-19 environment we're excited to be recommencing junior sport."
Hoy said the two clubs had to be thorough and do what was best for all involved.
"Naturally when something happens like that 10 minutes down the road (you have to be cautious)," he said.
"But the other thing too and the feedback from our parents is 'on Monday we put them all back on school buses and they're all going back to school and they're training anyway, so let's get them to play'.
"I think the protocols the league and clubs have put in place help minimise everything - play and leave, no access to change rooms."
The Saints and Seagulls plan to field a full complement of sides in both football and netball.
Games are at Koroit's Victoria Park with the under 18.5 football clash the showcase from 2.30pm.
