The Western District Health Service drive-thru clinic confirmed a fifth coronavirus case on Tuesday.
The man's partner was also tested on Tuesday and is awaiting her results.
The man who tested positive has been self-isolating in the Southern Grampians since he returned from overseas a little over a week ago, the health service said in a statement.
He was the fifth person to test positive to the coronavirus at the Hamilton clinic.
Six people have tested positive at Warrnambool testing clinics.
On Tuesday, it was revealed that the Warrnambool hospital had admitted its first coronavirus patient.
All others that have tested positive - who were aged in their 30s to 50s - have been recovering at home.
Cases are recorded on the Victorian Government's website each day as their local government area of residence, not where they are tested or self-isolating.
As of yesterday, there were four cases in Warrnambool and one each in Moyne, Corangamite and Glenelg.
Almost all are believed to be returned overseas travellers.
