A police media spokeswoman said officers were told a truck was travelling along Macarthur-Penshurst Road about 5.25pm when the driver lost control and the truck rolled.
The driver of the truck, believed to be a man in his 30s, died at the scene.
The man is the fourth person to die on south-west roads in 15 days.

EARLIER: The Penshurst-Macarthur Road has been closed by police following another serious motor vehicle collision on south-west roads.
VicTraffic is reporting the road is closed in both directions and are advising drivers to consider the Hamilton-Port Fairy Road to Hamilton and then taking the Hamilton Highway to Penshurst to avoid the accident scene.
The accident was reported at this evening, near the intersection of the Macarthur-Penshurst Road and Phillips Road at Warrabkook.
Emergency services are on scene, and the police will re-open the road when it is safe to do so.
