The B-double truck was travelling in an easterly direction on the Woolsthorpe-Heywood Road when it rolled onto its side just before midday on Monday.
Country Fire Authority District Five operations manager Richard Bourke said the truck was carrying about 60 steers, many of which were euthanised after the crash.
Mr Bourke said he did not believe any cattle would survive.
Police, CFA, SES and Agriculture Victoria vets are on the scene.
"The crews are working to extract the dead animals," Mr Bourke said.
"CFA were on site because when the truck was righted they had to be on site to make sure there was no fire."
He said crews would likely be on scene until 9pm Monday night.
An Ambulance Victoria spokeswoman said paramedics treated a man at the scene but he did not require transportation to hospital.
