Committee member Peter Carrucan said volunteers will plant trees and shrubs grown from seeds collected on the reserve.
"This is our second year running this project, last year we put in about 10,000 plants over four days and we did direct seeding as well," Mr Carrucan said.
"Volunteers will need to wear suitable clothes and footwear for the weather on the day, and there are leeches (on the property).
"All tree planting equipment will be provided, but they may also wish to bring their own gloves.
"We will provide free soup for morning tea when they arrive, plus a free lunch.
"People just need to bring a good attitude and a good appetite."
Established in 1990 the 254-hectare Pallisters Reserve property is home to a wide range of native flora and fauna. Local Indigenous seedlings to be planted on Sunday include tea tree, wattle, gums, grasses, sedges and tussocks.
"This is a great chance for people to see some real bush," Mr Carrucan said.
"We have over 130 bird species including brolgas, raptors, and parrots, two species of wallaby, Eastern grey kangaroos, echidnas and koalas on the property.
"There will be a bush walk to the Pallister block where we will point out different species along the way."
Mr Carrucan said the plan was to return the block to as close to original condition as possible.
"We will never get rid of all the weeds, and the animals and birds don't care if it is a weed if the plant suits their needs," he said.

People not wanting to travel to Orford on Sunday can still volunteer to plant trees as Reveg The Flume gets set for its 15th year.
Started by local surfers and Warrnambool Coastcare Landcare volunteers, it is Warrnambool's premier community tree planting event and is run in conjunction with National Tree Day.
Organiser Bruce Campbell said the community had transformed an area of introduced marram grass and drifting sand into a showcase piece of environmental work.
"This site demonstrates that we have the power to repair damaged environments," Mr Campbell said.
Volunteers should bring gloves and a small garden trowel if they have them. A free BBQ will be provided after the planting.
Pallisters Reserve Protecting Brolgas and Bandicoots community planting day runs from 10am-2pm at 457 Masons Road Orford.
Reveg The Flume runs from 10am-12pm on the corner of Merri Street, Warrnambool, and the Flume car park.
