Creek took its recruits tally to 19 when it signed three players just before round one.
Onballer Louis O'Connor and forward Joe Kenna will play on Saturday but half-back Jacob Lacy will miss this season with an anterior cruciate ligament injury.
O'Connor, 29, is returning to his junior club after time with Warrnambool in the Hampden league.
He said there was a great atmosphere at the Roo's Mack Oval.
"It's really positive, the vibes on the training track have been fantastic," he said.
"I think we would have had 40-plus (players) on Tuesday night."
Kenna, 23, has switched from Old Collegians.
"I thought I'd come and play with a couple of mates and see how we go," he said.
Lacy, 23, also from Old Collegians said he would assist coach Peter Timms while recovering from injury.
"I'm here to learn about the other side of footy, like coaching," he said.
"It should be good fun."
Creek have nine players playing their first game for the club on Saturday.
The other new faces are Liam Dwyer, Oliver Westwick and Caleb Bidmade (South Rovers), Ryan Bunney and Lukas Essenwanger (Koroit), Matthew Petherick (Old Collegians) and Caleb Templeton (East Warrnambool).
Timms said his recruits had earned their spot.
"The nine blokes coming in are coming into the side due to their hard work," he said.
"They've learnt the structure we've put in place and been solid during pre-season."
Timms said he had brought in so many players in the off-season to create an "accountable list".
"It will really put pressure on everyone," he said.
Creek will also have a couple of returning players with Timms revealing on Thursday that former coach and ruckman Heath Moloney was coming back from Darwin to play for the club.
Past player Billy McPhee will also return to the field on Saturday.
Timms said he was excited to see how his new-look side goes against the Tigers at the Merrivale Recreation Reserve.
"We're going to take the game on with a lot of run and carry," he said.
"We're a pretty quick side, hoping to get on the outside a lot.
"It will be hotly contested in the midfield."
Russells Creek is hoping to build on its ninth-place finish last year.
Timms, a former South Rovers three-time junior premiership coach and senior assistant, was appointed Creek's coach during the off-season.
