Marcus Herbert, 16, who will debut against Murray Bushrangers on Sunday, and older brother Liam, 18, will join a small group of players to line up alongside their siblings.
Talent manager Phil Partington said it was a rare and exciting feat for the brothers.
"We did a count out with brothers who have played and we worked out we have had 30 brothers that have played Rebels, which is an unusual fact," he said.
"But two brothers playing together in the one team that number would be less than 10. It's fantastic to see Marcus debuting with his brother in the side. It's also exciting for South Warrnambool and their family too."
Partington said Marcus and fellow debutant and Rooster teammate Fraser Marris, 17, had earned their spots.
"Marcus played with us in the under 16 development squad last year and has trained well over the pre-season and played well in trial our games," he said.
"He is a fit young man who can run and he deserves his spot as a 16-year-old.
"Fraser is similar. He has worked extremely hard over the off-season and he will come in and play that small forward role and he has shown he is a great pressure forward."
Warrnambool's Patrick Rea will also make his debut in the game.
The Herbert brothers and Marris will also play with South teammates Emmanuel Ajang, Jay Rantall and Isaac Thomas.
"To have six from South Warrnambool shows the strength of the junior program and how high ranked a club they and also how high ranked the league is with its juniors," Partington said.
The Hampden league will be well represented in this week's team with 13 players - including the seven Roosters - set to hit Bendigo's Queen Elizabeth Oval.
"Over half the team this week has come from the Hampden region and it shows how strong they are and it's something the league should hang its hat on," Partington said.
"It's super exciting that more boys are getting a chance to play in that (the NAB) league."
Hamilton Kangaroos' James Cleaver, Terang Mortlake's Isaac Wareham, Port Fairy's Josh Dwyer, Warrnambool's Mitch Burgess and Koroit's Connor Hinkley are the other five names in the team this weekend.
Fellow Kangaroo Shelby Nolte, 16, will make her debut in the Rebels under 18 girls encounter against the Western Jets.
