Portland police Sergeant Ray Hastings said the accident at 8.30am happened on the Myamyn-Condah Road, almost 50 kilometres north of Portland.
He said the single vehicle accident involved the driver losing control and hitting a tree about 8.30am.
"The vehicle has rolled and the car had been demolished. The 18-year-old male driver has been flown by air ambulance to the Royal Melbourne Hospital," he said.
"He has suspected head injuries and was in a serious but stable condition when he was put in the helicopter."
Sergeant Hastings said there was a large kangaroo dead on the road.
"The driver may have tried to go around the kangaroo and lost control. We won't know until we talk to the driver," he said.
