The two teams have agreed to a fixture swap for the match, and both will sit out during round 14 so the showdown can go ahead on Thursday, April 18.
It is believed to be the only match to be played on Good Friday eve across the state.
Heathmere lost the 2018 senior grand final to Heywood by 30 points, while Tyrendarra finished sixth on the ladder.
