Police and firefighters attended after the B-double rolled at the intersection of Koo Wee Rup Road and Manks Road, about 60 kilometres south-east of Melbourne, at 7.30am.
It’s believed the truck is a Boyles Livestock Transport vehicle, based at Naringal.
"Several cattle are being euthanised at the scene with assistance from the Department of Agriculture," Victoria Police said in a statement.
"The remaining cattle are being herded into nearby paddocks and the recovery operation is expected to take several hours."
The 42-year-old Warrnambool woman driving the truck was the only person in the vehicle and is not believed to be injured, police said.
The cause of the collision is still being investigated.
