Those flags include Timboon 1968, Heytesbury 1988, 1998 and Timboon 2008.
Vice president Brendan Hickey said it would be a significant day for the club.
"To tie in four premiership reunions in one day is pretty special," he said.
"A majority of people that played in them are all from the Timboon area and are locals that span across many generations.
"So it will be very exciting.”
The club is expecting to have on average 15 players from each premiership in attendance.
The four premierships will be the theme of the day, though current coach Mick Hunt said it was not all about the flags.
Hunt, who coached Heytesbury to premiership glory in 1988, said the event was a celebration of the entire club.
“The reunion is not just about the premierships,” he said.
“It’s also about the club and the fact of where we are now and where we were then.
“There are a lot of people that didn’t play in flags that did great things for the club.
“So that’s why it’s not just celebrating premierships.”
The festivities start as players and supporters, past and present, gather to watch the Timboon Demons in action.
After Saturday’s senior match against Dennington, the celebrations will continue with a dinner.
There will also be several interviews as well as footage being played of the various premierships.
Hickey, who is also the organiser of the past players and supporters day, said that the event looked set to stay on the calendar.
"Last year was the first year we kicked it off with the hope of it becoming an annual event,” he said.
"And we are confident and it could become that.
"For people that can't always get to the footy, this is the day not to be missed."
