It is believed a young woman driver had to be cut free from her vehicle following the accident, which happened about 8.20pm near Cudgee.
Warrnambool police said the woman was taken to Warrnambool Base Hospital with non-life-threatening injuries.
The crash, involving several head of cattle, blocked the highway in both directions until about 10pm and prompted a large-scale turn-out of police, ambulance, Country Fire Authority and State Emergency Service crews.
A ranger was also called to deal with the injured livestock.
Traffic banked up either side of the crash scene as the injured woman was treated in the car, which came to rest clear of the highway, and SES members worked to cut apart the vehicle for her to be lifted safely clear.
The badly damaged vehicle had to be towed away.
Large transports lined the sides of the highway while the accident scene was closed. Other traffic turned back or was diverted along other routes.
