The truck driver received minor injuries when the semi-trailer rolled over about 3.45am.
A police spokeswoman said the accident happened when the unloaded truck was travelling east.
The spokeswoman said the driver lost control and swerved to the wrong side of the road.
The truck rolled over a guard rail on the side of the highway and came to rest on the driver’s side door, the police spokeswoman said.
The driver was taken by ambulance to Colac hospital for treatment.
A heavy haulage vehicle was to be used to retrieve the semi-trailer and VicRoads detoured traffic along the Cobden-Stonyford Road.
