The Bloods' emphatic second term proved the catalyst for the 16.15 (111) to 11.9 (75) win.
They kicked seven unanswered goals in the second quarter, turning a one-point quarter-time deficit into a 42-point lead.
Port Fairy responded in the third quarter, kicking five goals to one to bridge the gap to 18 points at the final break.
But the Bloods had the answers.
Stephen Staunton kicked five goals for the winners and Jake Meade - one of Terang Mortlake's best in its second quarter burst - snared four.
Gary Robinson slotted four goals for the Seagulls, including the first two of the game.
Reigning premier Warrnambool thumped North Warrnambool Eagles in the other Saturday fixture at Reid Oval.
The Blues produced a 17.19 (121) to 7.9 (51) victory.
Skipper Jason Rowan booted eight goals as the Blues made it three straight wins to start their flag defence.
It was a fiery game, with Eagles onballer Andy McMeel sent off and reported.
Eagle Ben Mugavin and Blue John Paulin were also sent off for different incidents.
North Warrnambool Eagles were on the back foot early, losing Brendan Murfett and Luke Williams to pre-game injuries.
Lachlan Woodhams and Matt Brophy were called up to the senior side at late notice.
Koroit, Cobden and Portland were Anzac Day winners on Friday.
The Saints crushed South Warrnambool by 56 points, Cobden recorded an 18-point win against Camperdown and Portland notched a hard fought 17-point victory against Hamilton Kangaroos.
