He said a 32-year-old man driving a blue hatchback lost control of the vehicle east of Hamilton.
"The man driving the vehicle lost control and came to rest in a thicket of blackberries," Sergeant Habel said.
He said initial reports were that the driver and passenger were trapped in the vehicle but that wasn't the case.
Sergeant Habel said the driver and passenger were taken to Western District Health Service's Hamilton Base Hospital with non life-threatening injuries.
He said police were investigating whether fatigue was a factor in the accident.
"Hamilton police are keen to hear from members of the public who saw that vehicle travelling towards Hamilton at about that time," Sergeant Habel said.
An Ambulance Victoria spokeswoman said paramedics were called to a crash in Tarrington about 7.40am yesterday.
"A man in his 30s was taken to Hamilton Base Hospital in a stable condition with upper body injuries," the spokeswoman said.
"A woman in her 20s was taken to Hamilton Base Hospital in a stable condition with upper and lower body injuries."
He urged witnesses to contact Hamilton police on 5551 9100 or Crime Stoppers on 1800 333 000.
