The juniors will hit the court during the day on Saturday before the open and elite categories play in the evening.
CLB general manager Matt Hollard said there was plenty to be excited as fellow CLB club Sydney Supersonics had a team in the elite men's.
He added Aussie 3X3 representatives Tom Wright and Cooper Wilks were playing.
Hamilton's Joel Le Roy is playing in the elite grade with Samuel Brewer, James Cleaver and Josh Bourke.
Le Roy also stoked the Supersonics have entered a team.
"It's pretty big, they usually win the tournaments they play, they're a very good side," the 19-year-old said.
Le Roy, who also plays five-on-five domestic basketball, enjoys 3X3 because of the opportunities it presents.
He aspires to compete for the Rams' representative side when it gets the chance to play in a national league.
He and his mates won the under 18s when the first CLB 3X3 event came to Hamilton four years ago.
The teenager has played every year since and this year competed at 3X3 tournaments in Bendigo and Portland.
He said 3X3 was much faster than traditional basketball.
"You've got to be smart about how you play and shoot a lot," he said.
