Victoria police confirmed two Mount Richmond women, aged 16 and 43, died in a collision on the Portland Nelson Road.
The 16-year-old was driving a Holden Trax with the 43-year-old a passenger in the front of the car.
A 15-year-old female in the rear seat suffered serious injuries and was flown to the Royal Children's Hospital.
A spokeswoman confirmed to The Standard on Friday the teenager was in a stable condition.
The head-on collision occurred about 10.25am and the driver of a Jeep Cherokee, a 50-year-old man, sustained non-life-threatening injuries.
He was flown to the Royal Melbourne Hospital in a serious but stable condition.
Warrnambool police Sergeant Martin Flannery said the deaths would rock the Portland community.
"We really do feel for the family," Sergeant Flannery said.
"There is a human side to this. It rocks the community and emergency service workers are also affected."
Major Collision Investigation Unit detectives are investigating the circumstances of the crash.

CHRISTMAS EVE: Two people have died in a car crash near Portland on Christmas Eve.
Police are investigating after two women died following a two vehicle collision in Mount Richmond.
Emergency services responded to reports of a head on collision between a Jeep Cherokee and a Holden Trax on the Portland Nelson Road about 10.25am.
The driver and front seat passenger in the Holden died at the scene and a female child in the rear passenger seat sustained life-threatening injuries.
She has been airlifted to hospital.
The driver of the Jeep, a 50-year-old man sustained non life-threatening injuries and is currently assisting investigators with their enquiries.

The Portland-Nelson Road is closed in both directions through Mount Richmond.
The Department of Transport suggest motorists use the Princes Highway via Heywood and Winnap instead and allow extra time for their journey.
This closure is under Victoria Police control. The road will re-open when investigations are complete and it's safe to do so.
