Nathan snared 3-10 and made an unbeaten 38 and younger sibling Mark chimed in with 1-12 and made 49 not out as the Bulls overcame Dennington.
Brierly Christ-Church captain Matthew Love said the duo set the tone in the 10-wicket win.
The Bulls, who got Prahran's Nick King out for seven in his comeback game for the Dogs, restricted the home team to 91.
It included a five-run penalty for a slow over rate. They surpassed the total in 12 overs.
"We set it up with the ball, hit the right areas, fielded really well and made it hard for them to score," Love said.
"Then the Murphy brothers got together. They play T20 cricket beautifully, run really well between the wickets and make it hard for opposition captains and bowlers to set fields.
"They made it look convincing."
Love said he enjoyed the Thursday night twilight game concept.
It complements the Saturday one-day fixtures.
"It breaks up the week, six games in two-and-half weeks (for us) and because they're all worth six points, it makes it a little bit more important to get the wins," he said.
"It's not just hit-and-giggle."
In other games, Nestles celebrated captain Geoff 'Wizz' Williams' 200th game in style, scoring a 27-run win over Allansford-Panmure.
Williams made a half century in his milestone. Ben Boyd took 4-16 for the Gators.
The Factory posted 106 with the Gators all out in reply for 79 in the last over.
Woodford produced a 56-run win against Wesley-CBC with Jarryd Lewis claiming 3-10 in his first division one game of the year.
Captain Alex Jennings scored a half-century in Port Fairy's eight-wicket win against West Warrnambool.
The Panthers made 5-117 before Jennings, Jason Perera and Max Green, who made a quickfire 20-odd, steered Pirates home.
