The Department of Health and Human Services confirmed Warrnambool's local government area case numbers increased to six overall including one active overnight.
This is the first case recorded in the region for over 14 weeks.
It is not known where the person tested positive to the virus.
DHHS will conduct a thorough contact-tracing investigation of the active case.
Victoria has recorded 273 new cases of coronavirus since Saturday, with the total number of cases now at 3799.
Within the state, 22 of the new cases are linked to outbreaks, 25 were identified through routine testing and 226 are under investigation. No new cases have been detected in returned travellers in hotel quarantine.
There has been one death reported since yesterday, with a man in his 70s passing away in hospital overnight. To date, 24 people have died from coronavirus in Victoria.
More than 1,125,400 tests for the virus have been processed since the beginning of the pandemic.
