Chief executive Jackie Kelly said the introduction of testing came after a family of four holidaying in Port Fairy were confirmed to have the virus on Tuesday.
"This really came from our community," she said. "There's a lot of anxiousness about what happened over the weekend and we want to support the health of the Port Fairy community.
"We've had an increase in the number of people asking about testing and we understand there has been a lot of calls to the respiratory clinic in Warrnambool."
The short-term drive-through COVID-19 swabbing clinic is for residents of Port Fairy who are symptomatic to the virus.
The clinic will be open from 11am-2pm on Thursday and Friday.
Moyne Health Services will reassess continuing the clinic on Friday.
