The south-west patient returned a positive test today after presenting at Colac Area Health's designated COVID-19 test centre on Wednesday.
Colac Area Health chief Fiona Brew told the Colac Herald the patient was currently in self-isolation and that the hospital had informed the Department of Health and Human Services of the case.
DHHS will investigate the case further.
It brings the state's cases to 1365, an increase of three since yesterday.
The number of cases in the rest of the south-west remained unchanged as of Friday, with five cases in Warrnambool, two in Southern Grampians and one in Moyne, Corangamite and Glenelg.
