The woman was tested at Western District Health Service's drive-thru and was confirmed to have the virus on Wednesday.
Her partner tested positive on Tuesday after he returned from overseas a little over a week ago and has been self-isolating.
The couple are the first official Southern Grampians cases, with four other people who tested positive at the drive-thru either living interstate or in neighbouring shires.
There are also four cases in Warrnambool and one each in Moyne, Corangamite and Glenelg.
Almost all are believed to be returned overseas travellers.
