The four-player team of Donna Weller, Karon McKenzie, Mandy Dalton and Michelle Gristede edged out Warrnambool 1 by five shots.
The Marj Robinson Bowl - which is played in honour of a former Warrnambool Golf Club ladies captain, president, secretary and life member - is decided on the second day.
The victorious team is decided by who finishes with the highest aggregate stableford score.
Timboon finished with a score of 143 to beat the Warrnambool team of Di Goodall, Maida Gilbert, Rose Voigt and Deb Rix.
Last year's victorious club Terang finished in third with a score of 137. Kate Cornelissen and Judy Carmody were the only team members to return to the defend their crown.
It is the fourth time Timboon has won the annual tournament. The other three times were in 2002, 1997 and 1990.

Donna Weller, who is also a member of Warrnambool Golf Club, said it was an honour for the quartet to win the prestigious ladies tournament.
"Wow, how good was that for these girls to play so well and win here and a lot of them having only been playing golf for the last five or six years," she said.
"(We have) not really (played much golf together). Karon and I have gone away to play in bowls before but not Mandy or Michelle, who was playing in her second bowl.
"Going to different places and courses has really helped them and also playing golf more."
Weller, who has played in a number of Marj Robinson Bowls but had never won one, said the victory would boost the quartet heading into future tournaments.
"Beating the Warrnambool girls on their home course doesn't happen often," she said. "It's a great experience for the younger ones that haven't played in this and you just have to play your own game and it is what it is."
Mandy Dalton (B grade) and Karon McKenzie (C grade) also secured victories in the individual awards at the tournament.
Dalton finished with 35 points, edging out Warrnambool's Deb Fitzgibbon by one point. McKenzie (41 points) also took out her win by one point over Port Fairy's Kathleen Woodward.
Terang's Judy Carmody (41 points) secured the A grade victory with Warrnambool's Debbie Rix four points behind in second.
Dalton, Lorraine Wright (B grade), Ballarat's Julie Carr, Hepburn Springs' Bev Smith (A grade), Warrnambool's Maida Gilbert and Kim Dummett (C grade) were the closest to the pin on either the ninth or 13th holes.
Over 200 competitors - 120 on Monday and 107 on the final day - played across four-ball better-ball and team matches in the two-day tournament. Twenty-two clubs were represented.
