Portland would have under 14 and under 16 sides while Hamilton Kangaroos would add an under 14 team to the Hampden Junior Competition.
President Tim Mason said he was thrilled Portland and Hamilton junior sides could join the league next season.
"We are very pleased to welcome football sides from both Portland and Hamilton for the 2020 season," he said.
"We, as a board, believe that the introduction of these teams is only going to further strengthen our league and the quality of the junior competition.
"Allowing both clubs to enter these teams will further strengthen the football communities in their respective towns by allowing more junior players to test themselves within the Hampden League."
The league will present draft fixtures for the senior and junior competitions at its 2019 annual general meeting.
