The Seagulls picked Matt Sully, Johnny Bell and NAB League-listed Josh Dwyer to face the Roosters at Koroit's Victoria Park.
Boom recruit Tom Sullivan and Andrew Lowe will both miss with injuries with the club "not taking any risks".
Jake Bartlett, who has played 11 senior games in 2019, was the unlucky player to be omitted.
Port Fairy assistant coach Winis Imbi said the Seagulls were thrilled to add former VFL team of the year defender Sully and the versatile Bell and Dwyer.
"The good thing is those players are multi-positional," he said.
"People watching us play know we utilise Matt wherever we feels he gives us an edge, Johnny can play back but has kicked over 100 goals at South West District level as well.
"Josh covers the ground exceptionally well and can play back, wing and half-forward."
Imbi said there was "genuine disappointment for young Jake Bartlett".
"He's impressed for us all year and had really good growth," he said.
"We do feel for him but hopefully there is a bit more left in us and he can train hard (and push to regain his spot)."
Port Fairy's opponent, South Warrnambool, named an extended squad and will train again on Friday night before finalising its team.
But Roosters coach Mat Battistello said brothers Liam and Marcus Herbert, who are available following Rebels' commitments, would play.
AFL draft prospect Jay Rantall was not considered after a heavy workload as he prepares for October's combine in Melbourne.
"We understand he probably wants to wear two jumpers but at the end of the day, the most important thing for Jay is to present himself really well for draft combine so he won't play this week," Battistello said.
Warrnambool and Koroit both made two changes for their qualifying final at Hamilton's Melville Oval on Saturday.
The Blues added midfield grunt with Damien McCorkell returning from a knee complaint and the skilful Mitch Burgess returns from NAB League duties.
Youngsters Wil Fleming and Tom Jenkins were omitted.
Premiership midfielders James Gow and Liam Hoy were added to the Saints' line-up with Frazer Robb and Lachlan Cairns missing out.
