Premiership defender Daniel Threlfall will boost the back line while Jesse Dalton and Aaron Searle will provide serious firepower up forward.
Dalton, who returns from a knee injury, hasn't played since May.
He kicked 11 goals and appeared in the best three out of four times before he was brought down by the injury.
Co-coach Shane Threlfall said Josh Irving, Dylan Willsher and Aaron Smith were the "really unlucky" players making way for the star trio.
The Blues mentor is anticipating a blockbuster because South Rovers desperately needs to win if it is to have any hope of finals.
"We're expecting them to come out all fired up," he said.
South Rovers will need to pull off a massive upset and Dechlan Picken is back from hamstring tightness to help the cause.
Lions Eamon Dowd and Thomas White are both unavailable.
Kolora-Noorat star midfielder Scott Judd will play his 100th match for the club.

Dennington's Ben Mulcahy, 16, will make his senior debut against the Power.
Coach Mick Phillips said it was a reward for his persistence.
"He trained all pre-season and he's improved a lot since he first came to training," he said.
Injuries once again trouble the Dogs with John Vincent (knee complaint), Tully Allwood (adductor), Dillon Paton (foot) and Trent Torney (calf) ruled out.
Russells Creek mentor Peter Timms has shuffled plenty of magnets with six changes.
Liam Antonio, fresh from his trip to London, will slot straight back in at centre half-forward.
Creek's spine will also welcome Harry Hall at centre half-back who returns for his second game of the year due to an ankle injury.
"We've really missed him," Timms said.
East Warrnambool will meet Creek with Hayden Harradine and Liam Feros-Collins back in the side.
But it will be missing Marcus Clarke.
Panmure, a team chasing finals, welcomes key-position player Jaymen Fletcher back for his first senior match of the season.
Josh Parsons (thumb) and Thomas Wright (back complaint) also return.
Timboon Demons has a debutant with Anthony Glennen to join Marcus Hickey's line-up.
