Police believe the truck was travelling in front of the bus when its trailers dislodged and blocked the highway in both directions, causing the crash and trapping the driver, who died.
The accident happened near Pimpinio about 2am on Thursday, with at least eight people injured, Ambulance Victoria said.
Pimpinio is about 15 kilometres north of Horsham on the road to Dimboola.
Paramedics were treating the eight victims for various injuries, and two were flown by air ambulance plane to Melbourne.
The other six were taken by road to Wimmera Health Care in Horsham.
More than 30 other people from the bus will also be taken to Wimmera Health Care for assessment.
The highway is closed in both directions as a result of the crash, with diversions in place and detectives at the scene.
The diversions are expected in place for much of the day.
