The ex-Fremantle and Gold Coast ball magnet, whose last game at the elite level was just 10 months ago, is listed with VFL club Werribee.
He picked the Eagles as his feeder club - brother Herb used to play there - and was available for selection due to the Tigers' bye.
Eagles coach Adam Dowie said the club was thrilled to have Barlow, who averaged 25 disposals across his nine-year AFL career, for its clash against Hamilton Kangaroos at Bushfield.
"I spoke to him yesterday and I thought he'd be keen for a week off but he is excited about playing," he said.
"He's got the family connections and everyone is pretty up and about about him playing. It's something he's been hoping to do for a while."
North Warrnambool Eagles also brought in teenagers Harrison Sinclair, Tom Keast and Alex Jennings.
Pacy midfielder Sinclair will play his first senior game.
Camperdown had its own selection boost, welcoming the grandson of former Geelong player Stewart Lord into the fold.
Will Lord has joined the Magpies from Geelong league club St Albans after moving to the area for work. The midfielder is also on VFL club Footscray's development list.
Pies co-coach Neville Swayn said the new recruit had strong ties to the club.
"His grandfather (Stewart) coached Camperdown to two flags," he said.
Teenager Sid Bradshaw will make his senior debut for the Pies.
"He's a left-footer and a good ball user," Swayn said.
The Magpies' opponent, Portland, has made five changes with coach Tom Sharp (general soreness), midfielder Aaron Shepherd (unavailable), teenager Harris Jennings (Vic Country commitments), Harry McIntyre (cut hand) and Damon Huppatz (omitted) going out.
The versatile Jake Wilson headlines the Tigers' inclusions.
Warrnambool has unveiled a debutant for its clash with arch rival South Warrnambool at Reid Oval with teenager Zacc Dwyer to play on a wing.
"He's pretty aggressive, a good user of the ball and a good tackler in the under 18s," Blues coach Matt O'Brien said.
"He's fast so hopefully we get the footy in his hands a little bit."
Defender Matt Colbert comes in for his first senior game of the year after battling injuries.
"He's a very tight defender, reliable, spoils the ball well and is a good tackler too," O'Brien said.
Terang Mortlake and Cobden have both made multiple changes for their clash.
The Bloods welcome back Steve Staunton, who will slot into the ruck, Sam Crawley, Dylan Jones and Harry Williams.
The Bombers brought in Jack McGlade, Ben Berry, Jia Anderson, Nick Nieuwenhuizen and Patty Smith for the injured Tom Watson (hamstring), Ned Dalziel (knee), Mitch Reed (concussion), Tomas Lindquist (calf) and the suspended Paul Pekin.
Cobden coach Adam Courtney said the club was thrilled to welcome impressive teenager Smith back in after a series of injury concerns.
"He's the future midfielder of the football club," he said.
"He's an exciting talent and it's great to have him in the side."
Port Fairy will be without Sandy Robinson (unavailable) and Sam McCartney (injured) for its clash against fierce rival Koroit at Gardens Oval.
Joe Forrest will debut for the Seagulls and defender Dylan Herbertson returns to the side.
The Saints welcome back Sam Dobson, Rhys Raymond and Daniel Harrington from injury.
Defender Harrington has spent a long stint on the sidelines with multiple injuries.
George Serra, who debuted last week, is out with a hamstring strain and Daniel Roache (ankle) and Todd White (Vic Country) also miss.
Meanwhile, eight Hampden footballers - six current and two past - are in contention to play for Vic Country against Victorian Amateur Football Association next month.
Port Fairy trio Dan Nicholson, Tom Sullivan and Kaine Mercovich and Koroit trio Ben Dobson, Sam Dobson and Tom Couch were picked in the initial 58-player squad for the Brian Moloney Cup on July 13.
Ex-South Warrnambool utility Louis Herbert (South Barwon) and former Terang Mortlake midfielder Xavier Moloney (Swan Hill) are also chances to play in the annual grudge match.
Former St Kilda defender and Richmond coach Danny Frawley will coach the Vic Country side.
Four training sessions will be run in the lead-up to the match at Carlton's Ikon Park with the squad to be trimmed to 22.
