Tom Sullivan will miss the clash after failing to recover from a corked thigh he sustained in the Jonathan Brown-led Hampden league side's AFL Victoria Community Championships win over Ballarat last weekend.
The 24-year-old, who was one of the Bottle Greens' best players after slotting three goals in a half before his injury, has played every game for the Seagulls this season as well as booting seven majors.
MORE FOOTBALL:
John Bell (unavailable) and Lochie Gunning (groin) will also miss the clash, while Greater Western Rebels-listed player Josh Dwyer returns alongside Kurt Smith and Blake Carroll.
Seagulls assistant coach Winis Imbi said he and the coaching panel took the conservative approach with Sullivan and Gunning.
"Tom is one of our marquee players and he has his own standards that he likes to be at. He is a terrific runner and when you lose one of the strengths of your game it compromises your standard," he said.
"We weren't prepared to risk him as it's only a corky but it was severe enough that he didn't play in last quarter last weekend."
Imbi said the Seagulls would back their system to cover the side's losses against the Blues, who go in unchanged for the second time this season.
Meanwhile, Koroit will welcome back a host of experience when captain Brett Harrington (hamstring), forward Will Couch (suspension) and defender Damien O'Connor (hamstring) return to the side.
Saints coach Chris McLaren named an extended bench as he waits to see how some of his stars pull up from last weekends interleague.
North Warrnambool Eagles have also named an extended bench with Billie Smedts and Tim O'Brien returning to the side, while Jye O'Brien is the only omission so far.
South Warrnambool coach Mat Battistello has made a host of changes with two debutants coming into the Roosters' starting 21 for Saturday's clash against Terang Mortlake.
Sixteen-year-old Archie Stevens and Rebels-listed player Emmanuel Ajang will make their senior debuts.
MORE FOOTBALL:
Ajang's GWV teammates Marcus Herbert, Fraser Marris and Isaac Thomas also return alongside co-captain Nick Thompson, Paddy Anderson and Brayden Beks.
Tim Ryan (finger), Brad Miller (managed), Tom Wilson (hand), Tate Porter (Merrivale) and Timothy Brown (away) are forced charges, while Heath Brennan, Tait Niddrie and Jed Henderson were omitted.
The Bloods made one forced change with Tyson Densley ruled out with a quad injury. He will be replaced by Matt Baxter, who returns from injury.
Portland will also unearth its own debutant when it faces Cobden on Saturday night. Toby Jennings will line up alongside his brother Harris for the first time in the Tiger's senior side.
Jennings will be joined by the returning Ben Malcolm (soreness), Connor Giddings (Rebels) and Toby Oakley.
Cobden have made just the one change with Thomas Watson coming in for Billy McGlade.
Camperdown welcomes back Fraser Lucas from suspension along with Luke Clarke, while also introudcing a debutant in James Byrne. The Magpies opponents, Hamilton, go in unchanged.
