The Warrnambool Wolves and Corangamite Lions senior teams will go head-to-head from 7pm in a round one grand final rematch on Saturday night for the history-making fixture.
Wolves president Brett Gasper said he asked Lions counterpart Simon Gleeson if his club's top side would like to play its opening match in a home-away-from-home situation.
"They allowed us to have it as we both wanted to make the best start to season for the league as we could," he said.
The night will begin with the under 12s kicking off at 4pm, which will be shortly followed by the under 14s at 5.30pm before the senior men battle it out.
The last time the two teams met Corangamite claimed the title with a 3-2 victory over the league winners in the 2018 grand final.
Since then the competition has also expanded with Stawell Pioneers starting as a club and joining during the off-season.
Lions player Jamie Norton and Wolves striker Josh Bateman both said they were eager to tackle the night game.
"It's pretty good of the Wolves to host it. They have good facilities and we are feeling pretty confident," Norton said.
"There is a lot of young talent and some new faces coming through. I think there are four or five new players that haven't played in our senior side."
Bateman said the Wolves were prepared for another challenge at the league-premiership double.
"We are excited for the game under lights for the first time for the league and we are looking pretty strong," the 26-year-old said.
"We have a good squad going and had some good pre-season games and hopefully we can put that on the pitch come Saturday night."
Norton said the rivalry between the two clubs would continue to grow this season.
The 30-year-old said the Pioneers introduction into the league would be a new and welcomed challenge for the competition.
