The 16-year-old played at number seven and beat Commonwealth's Caleb Bovalina in a tight Victorian Pennant division one encounter at Yarra Yarra.
MacKenzie was one up going into the final hole and held on as he and Bovalina finished with birdies.
Manager of Kingston Heath pennant Mark Tucker said the youngster played a major role in the victory.
"It was a significant win for us," he said.
"He set the path for us to win the day."
"His golf is developing really well."
Tucker confirmed that MacKenzie would remain in the senior team this Sunday.
Tucker said MacKenzie earned his spot in the top team after a string of victories in division one colts.
Will's dad Brian said his son did well to get over the line.
"It was a fairly tight match," he said.
"It was a good effort because they are under a bit of pressure."
MacKenzie has played Kingston Heath colts for the past three years and stepped up for one senior appearance last year.
MacKenzie, who is based in Warrnambool, travels to Melbourne every weekend for the seven rounds of pennant.
Meanwhile, Hamilton is hoping for two in row after it defeated Port Fairy in a close contest in the first round of the Western District Golf Association men's division one pennnant.
Hamilton captain Malcolm Groves said it was a significant milestone for his team.
"It's the first time we've beaten them in the new district," he said.
He said his team's number six and seven - Daniel Battista and Jason Chandler - got them off to a strong start.
"They had wins that both went down to the 18th hole," he said.
Groves said the day was decided in the final match when Mat Walkenhorst birdied the last to finish two up against Matt Cameron.
Groves said his side had turned its attention to this Sunday when they face another powerhouse, Warrnambool 1, at Port Fairy.
The Hamilton captain said he hoped his side brought its best and claimed two victories on the trot.
"It would be a great start if we could," he said.
