Sinclair shot 166, to sit six ahead of Lauren Higgins and Marion Venn who are each on 172 with one round to play.
The 10-time winner had a one-shot lead after the first round but hit 82 off the stick in the second to surge ahead.
Ladies club captain Higgins said she hoped a big crowd would come and watch the final round of the championship next Wednesday.
"We've had nice weather for both rounds, it's been favourable," she said.
"The course is in amazing condition."
Higgins hasn't won the coveted title before while Venn lifted the trophy in 2008.
Not far behind the top three is 2017 champion Di Robinson on 178.
The B grade competition has a thrilling contest at the top.
Rosemary Walters (189) is leading by one shot over Beryl Drake (190).
Karen Jenkins is next best with 195.
Walters has also taken out the Elsie Watson trophy which is for having the best nett score in the first two rounds of the championship.
Walters shot a nett score of 72 in first round and 71 in round two for a total of 143.
Higgins said the award was just for the first two rounds of the championship.
"It's quite a prestigious trophy," she said.
The C grade championship is also set for an exciting finish next Wednesday.
Ann Glennon (217) is hot on the heels of Rose Voigt who has shot 216.
