Officers have been told a B-double was travelling along Henty Highway just after 3am.
It is believed the prime mover veered off the road and crashed into a tree.
Emergency services attended the scene, but the truck driver had sustained fatal injuries.
Police are yet to establish the exact circumstances surrounding the crash and the investigation remains ongoing.
