With the 12-hour festival at Suffoir Vineyard set to reach temperatures in the high 30s on Saturday, festival secretary Michelle Badenhorst said being sunsmart was priority.
"We've got some new measures in place to help fight off the heat," she said.
"We've added extra sunscreen stations, water stations and we'll have sprinklers every hour."
Mrs Badenhorst said additional marquees had been donated by The Grampians and Moyne Shires.
Now in its fourth year, Music in the Vines is the brainchild of a group of Macarthur residents determined to increase tourism.
"We came together to celebrate what the local area has to offer and put our town on the map," Mrs Badenhorst said. "The vineyard seemed the perfect setting."
The festival runs from 11am to 11pm and attracts musical talent from across the region.
The not-for-profit event distributes proceeds among surrounding community groups.
"Last year was the first year we made a profit," Mrs Badenhorst said.
"We were able to help the Macarthur pool purchase a new slide.
"As soon as we have enough in the kitty we look at which community groups need our help."
This year's line up includes Woodey Pitney, Zeon, Cooper Lower, Flynn Gurry, Marcia Howard and The Still Trees.
Free shuttle buses will transport festival-goers from Warrnambool, Portland, Port Fairy and Hamilton.
