He failed to negotiate a bend, his vehicle nose-dived and flipped several times.
The driver was ejected from his vehicle.
Police will make inquiries into whether he was wearing a seatbelt.
Saturday, 11am: The Stonyford-Cobden Road is expected to be reopened within the next hour after a fatal crash in the early hours of Saturday morning.
Acting Sergeant Don Camilleri, of Camperdown police, said the 40-year-old south-west man was on his way home when his vehicle rolled three times on a bend in the road.
Police were notified about the accident about 4am when a passing truck driver came across the scene, Acting Sergeant Camilleri said.
The accident happened on the Stonyford-Cobden Road about 200 metres south of Koallah Road in South Purrumbete.
Earlier: The male driver of a car died after his vehicle rolled a number of times on the Cobden-Stonyford Road on Saturday.
A police media spokesperson said it was believed the vehicle was travelling along Cobden-Stonyford Road about 4am when it lost control at Stonyford and rolled.
The male driver and sole occupant, who is yet to be formally identified, died at the scene.
The exact circumstances surrounding the crash are yet to be established and the investigation is ongoing.
The police said road closures were expected to remain in place for some time.
