The 663 tonne superload is the largest seen on Victorian roads in recent years, and is being transported from the Port of Geelong to Haunted Gully at Lismore.
It departed at 11pm on Monday and stopped at the Waurn Ponds rest stop on Princes Highway between 5am and 11pm on Tuesday.
Overnight it passed through Winchelsea.
The load is currently blocking the Colac-Ballarat Road as it continues the second leg of the journey to Swan Marsh.
VicRoads says motorists should seek an alternative route and allow extra time.
The massive vehicle is 115 metres long, 5.12 metres wide and 5.5 metres high. It is being haukled by four four trucks in an operation spanning three nights to reduce the impact on the road network and alleviate congestion for drivers.
