Detective Senior Constable Rachael McMahon, of the Warrnambool police crime crime investigation unit, said a van was located by police after a single-vehicle accident on the Hopkins Falls Road at 12.45am Monday.
The driver was not injured and picked up by another motorist.
The van was seized by police, forensically analysed and a commercial quantity of cannabis was found in the rear.
"Two men have been assisting police with our inquiries," Detective Senior Constable McMahon said.
"The cannabis was also seized and investigations are continuing," she said.
The men are expected to appear in the Warrnambool Magistrates Court on Tuesday for bail/remand hearings.
