Warrnambool police Sergeant Tracie Kelly said a motorist who spotted the cow had pulled over on the side of the highway to flash their lights.
But a second driver didn’t see the animal and collided with it about 3am.
The cow died at the scene.
Sergeant Kelly said the animal was still located on the side of the highway.
“Council will be removing the cow today,” she said.
The driver was taken to the Warrnambool Base Hospital.
