Hamilton police Sergeant Michael Vaughan said the single-vehicle accident happened in the Strathdownie district about 4.30am.
He said the Mount Gambier driver lost control and ran into a tree, suffering non-life-threatening injuries.
The driver was transported by ambulance to the Mount Gambier hospital.
His car received extensive damage.
"Fatigue is believed to be a significant factor in the accident," Sergeant Vaughan said.
"We would take this opportunity to warn all drivers to drive to the road conditions, plan your trip and take a break if you feel tired," he said.
