Hussey will leave the country as he heads off travelling overseas despite the Roosters being set to play finals.
Coach Mat Buck said he hoped they could cover his absence.
“We knew he was leaving very early in the year and we have planned for it over the last five or six games,” he said.
“We have made adjustments so we hope the impact isn’t too severe, but it is hard to replace a James Hussey but we will see how we go.
“It’s an improvement across the board from everybody to fill that attacking void that James leaves.”
Two young Roosters are set for greater opportunities following his departure.
“Ethan Harvey and Ben Annett will have more of a chance to play across the ball,” he said.
“I am excited to watch them play higher minutes and see how great they can be.”
The Tigers head into the clash without Toby Oakley and Tylar Watts but welcome back Connor Giddings from the Greater Western Victoria Rebels.
Terang Mortlake experienced a mixed night at selection on Thursday with Alex Moloney coming back in along with Dylan Wareham.
Coach Michael Sargeant however confirmed Joel Crawley would miss the match with Warrnambool due to a back complaint.
“Alex is a big in for us, we have missed him the last couple of weeks so hopefully he can have an impact, but we aren’t putting great expectations on him,” he said.
“Joel got spasms in his back about five minutes before we ran out last Saturday so he is out and we will have to wait for them to settle down before we know more.”
Camperdown has named a four-man bench, but is keeping its cards close to its chest.
Coach Phil Carse said a couple of players would be assessed before selecting a final squad.
Port Fairy will be without the in-form Kaine Mercovich but bring in Xavier McCartney.
