Detective Senior Constable Richard Hughes, of the Warrnambool police crime investigation unit, said the 26-year-old victim had been staying at the hotel while working in Warrnambool.
"At about 5.30am the victim left the hotel via the Fairy Street entrance and went to the car park next to the hotel, with the intention of going for a coffee," he said.
"He states that an unknown man, wearing a hoodie, was standing in the shadows smoking a cigarette.
"The offender asked the victim for the time and when the victim looked at his mobile phone, the offender produced what is believed to be a steak knife and demanded the victim's keys."
Detective Senior Constable Hughes said the victim immediately jumped in his car and drove off.
"He returned a short time later, after collecting a work colleague, however, the male offender could not be found near the hotel," he said.
"The victim then went to work and contacted police at 8.20am."
The detective said he had contacted the Cally Hotel and security camera footage was being obtained to try and identify the offender.
He requested anyone with information contact the Warrnambool police station or Crime Stoppers on 1800 333 000.
