A driver aged in his 40s was treated at the scene just after 9am when his b-double truck jack-knifed near the intersection of Barwon Terrace.
Winchelsea police told The Standard the livestock truck had departed Warrnambool earlier in the morning and was returning to Pakenham when the incident occurred.
Traffic was diverted via Deans Marsh-Winchelsea Road and Cape Otway Road while CFA and SES members secured the scene.
Meanwhile, Portland police are seeking information after a car smashed into a fence in Edgar Street on Wednesday night.
A police spokesman said that about 7.30pm, a green Holden sedan was “hooning around” before it crashed into a fence.
The spokesman said a number of people in the car fled the scene, leaving the engine running.
“There was evidence in the car that showed someone was injured,” he said.
“Anyone with information is encouraged to call Crime Stoppers or Portland police.”
